How do I deploy my project in visual studio 2010 express edition? I dont see any setup and deployment template

Only ClickOnce deployment is available to the Express Editions.

but with clickonce is it possible to use the application on different computers?

Something to note, if you're an American, French, British, or Swiss college student attending a public college, and have a .edu email from that school, you more likely than not would have the ability to get a dreamspark (just Google dreamspark) account, which would entitle you to download Vis Studio Profession 2010 free, and more importantly legally. A lot of schools spend a bit of money to allow Microsoft to do that, so if you are a college students to do this, so if you are a student, you should at least try and see if you qualify.
