Human operator intelligence. Don't open attachments or use links in emails. Don't troll porn, torrent, LimeWire sites. Don't install hacked/pirate software. Don't install those free animated screen savers. Blah blah.
None of them and all of them. They all have their strengths, weaknesses and targeted audience.
An enterprise level A.V. application might excel at using their own server for only one copy of the definitions so each and every 5,000 workstations in the building don't have to update themselves individually for example.
Personally, I use AVG. But I also have my working OS as a VMware virtual machine running on top of the host OS to further isolate it from prying hacker eyes.

Emsisoft has the highest detection rates for malware and especially trojans. It provides anti-virus too but should not be used as a main anti-virus (imo)
I would suggest using AVG or Avast (free) with their anti-virus active protection.
Then Emsisoft for malware/trojan checks every couple of weeks.
MalwareBytes is also great for malware but Emsisoft's detection rates have impressed me. We found over 20 trojans on a computer already running Sophos antivirus with it the other day.

Kaspersky Antivirus 10 - no problem with it, detects anything it needs to detect.
