I've got a CD with approximately 5,000 files on it, and on one computer in the office it loads everything perfectly fine, but then on another computer it always tells me that one particular file is corrupt and cannot be accessed.
What could cause this?

Faulty CD/DVD drive? Can't really say when you only tested it on two machines, though. Try testing on some more and see how it goes.

Thanks for the recommendation, I will try it on some more machines. (For the record, no other discs have this issue, it's just one particular disc out of a series of over 200 that works fine on the first machine, but not on the second).

Dirty lazer lens. It's difficult to see the lens on a desktop, where as a laptop spits them out & they are easily viewable.
